Cefpodoxime Proxetil is a third-generation cephalosporin antibiotic, this is a prodrug absorbed from the GIT and to obtain cefpodoxime ,which is its active metabolite, de-esterification takes place.

Ofloxacin is a fluoroquinolone class of antibiotics and treats a wide range of bacterias. It inhibits the growth of bacteria by hindering its reproduction and repair of its genetic material i.e. DNA.

PHARMACOLOGY

CEFPODOXIME

MECHANISM OF ACTION: – The bacterial cell wall synthesis is interrupted. The cell wall synthesis is inhibited via the active metabolite of Cefpodoxime which has high affinity towards penicillin binding protein, this bonding interrupts the production of peptidoglycan which is the main constituent of bacterial cell walls. Thus the cell wall formation gets hindered and bacterial growth ceases.

PHARMACOKINETICS: –

• ABSORPTION: oral administration of the 100mg dose results into absorption of the drug.
• PROTEIN BINDING: – Less than 30% of binding is observed in serum as well as in the plasma.
• ELIMINATION: – 29-33% of Cefpodoxime is excreted through urine within 12 hours of administration.

OFLOXACIN

MECHANISM OF ACTION: – Being a broad-spectrum antibiotic it is active against both Gram-positive and Gram-negative bacteria. It inhibits the function of DNA gyrase, which is responsible for prohibiting the replication of DNA, thereby inhibiting bacterial cell division.

PHARMACOKINETICS: –

• ABSORPTION: – It has been found that approximately 98% is absorbed in the body when administered in tablet form. The concentration of serum reaches to a maximum in one to two hours after an oral dose.
• ELIMINATION: – After absorption an unchanged amount of orally administered drug is eliminated via kidneys. It takes about 48 hours to excrete about 65 to 80% of ofloxacin toget out through renal excretion. Another 8% of the drug concentration is eliminated in the feaces.

• for Pharyngitis/tonsilitis
• Community-acquired pneumonia
• Acute bacterial exacerbation of chronic bronchitis
• Acute, uncomplicated urethral and cervical gonorrhea
• Acute, uncomplicated ano-rectal infections in women
• Uncomplicated skin and skin structure infections
• Acute maxillary sinusitis
• Uncomplicated urinary tract infections (cystitis)
